Protein Expression Purification

Protein expression purification is a method for producing a target protein in a biological host and isolating it from other cellular components for study or use. Researchers introduce the gene encoding the protein into cells such as bacteria, yeast, or mammalian cells, induce expression under controlled conditions, and recover the product through cell lysis or secretion followed by techniques such as affinity, ion-exchange, or size-exclusion chromatography. In immunology and infection research, purified antigens, antibodies, and pathogen-associated proteins support immunoassays, structural studies, vaccine development, and diagnostic testing. Protein quality, folding, purity, and biological activity directly influence the reliability of downstream experiments.

In this protocol we demonstrate the expression, solubilization, and purification of a recombinantly expressed membrane protein, MexB, as a soluble protein detergent complex. MexB is a multidrug resistance membrane transporter from the opportunistic bacterial pathogen Pseudomonas aeruginosa.
