Rhesus Macaque Pbmc

Rhesus macaque peripheral blood mononuclear cells (PBMCs) are immune cells isolated from rhesus macaque blood to study cellular and molecular responses in a nonhuman primate model. They include lymphocytes and monocytes, which can be separated from whole blood by density-gradient centrifugation because these cells have lower density than erythrocytes and granulocytes. Researchers use rhesus macaque PBMCs to characterize T-cell and B-cell activity, analyze cytokine responses, and monitor immune changes during infection or vaccination. Their close physiological and immunological relationship to humans makes them valuable for evaluating disease mechanisms, vaccine candidates, and immune-based interventions before clinical studies.

We demonstrate the utility of multicolor flow cytometry for detailed phenotypic and functional characterization of total as well as memory subsets of CD4+ and CD8+ T cells in rhesus macaques, the ideal model for HIV/AIDS vaccine studies.

In this article, we describe isolation of rhesus macaque derived adipose-derived stem cells (ADSCs) using an enzymatic tissue digestion protocol. Next, we describe ADSC proliferation which includes cell detachment, counting and plating. Lastly, ADSC differentiation is described using specific adipogenic inducing agents. Additionally, we describe staining techniques to confirm differentiation.

Detection of Rhesus Factor in Human Blood Samples using Magnetic Levitation

This video demonstrates the detection of the Rhesus (Rh) factor in blood samples using a magnetic levitation instrument. The magnetic field aligns and levitates beads and blood cells based on density. Treating the samples with anti-Rh antibodies results in intermediate-density bead-cell complexes' formation, allowing for the differentiation of Rh-positive and Rh-negative red blood cells.

This article is useful not only to the field of HIV immunology, but also to other areas of biomedical research where CD8+ T cell responses are known to affect disease outcome.
