Tritiated Thymidine Incorporation

Tritiated thymidine incorporation is a radiometric assay that measures DNA synthesis as an indicator of cell proliferation. When cells enter S phase, they take up radiolabeled thymidine, incorporate it into newly formed DNA, and produce a signal proportional to proliferative activity; researchers typically quantify this signal by measuring radioactivity. In immunology and infection research, the method assesses lymphocyte responses to antigens or mitogens and evaluates how pathogens, immune factors, or treatments alter cell division. Although newer nonradioactive assays are available, tritiated thymidine incorporation remains a sensitive reference method for comparing proliferative responses in cultured cells.

An imaging-based method is described that can be used to identify S-phase and analyze cell cycle dynamics in the C. elegans hermaphrodite germline using the thymidine analog EdU. This method requires no transgenes and is compatible with immunofluorescent staining.

We have derived a strategy to detect sequential incorporation of thymidine analogues (CldU and IdU) into tissues of adult mice to quantify two successive rounds of cell division. This strategy is useful to detect cell turnover of long-lived tissues, oncogenic transformation, or transit-amplifying cells.

Bioinspired Soft Robot with Incorporated Microelectrodes

A bioinspired scaffold is fabricated by a soft photolithography technique using mechanically robust and electrically conductive hydrogels. The micropatterned hydrogels provide directional cardiomyocyte cell alignment, resulting in a tailored direction of actuation. Flexible microelectrodes are also integrated into the scaffold to bring electrical controllability for a self-actuating cardiac tissue.
