Cancer Signal Transduction

Cancer signal transduction is the study of how molecular signals move through cancer cells to regulate abnormal growth, survival, invasion, and treatment response. These pathways begin when extracellular cues or genetic alterations activate receptors and intracellular proteins, triggering phosphorylation cascades and transcriptional changes that can disrupt cell-cycle control, apoptosis, angiogenesis, or immune recognition. Key networks such as RAS–MAPK, PI3K–AKT–mTOR, and JAK–STAT help researchers explain tumor behavior and identify therapeutic targets. Mapping these signaling mechanisms supports biomarker development, patient stratification, and the design of targeted treatments, while also clarifying how tumors evolve drug resistance.

We present guidelines for developing synthetic 'chemical transducers' that can induce communication between naturally unrelated proteins. In addition, detailed protocols are presented for synthesizing and testing a specific 'transducer' that enables a growth factor to activate a detoxifying enzyme and consequently, to regulate the cleavage of an anticancer prodrug.
