Epicardial Injection

Epicardial injection is a procedure that delivers therapeutic agents directly to the outer surface of the heart, providing localized treatment of underlying cardiac tissue. During the procedure, a needle deposits drugs, biomaterials, cells, or gene therapy vectors into or beneath the epicardium, where they can diffuse into the myocardium and limit systemic exposure. Researchers use this approach to study cardiac regeneration, myocardial infarction repair, drug delivery, and treatment of localized cardiac disease. Its effectiveness depends on accurate targeting, tissue retention, biocompatibility, and safe access to the beating heart, making it an important technique in translational cardiovascular medicine.

Epicardial Outgrowth Culture Assay and Ex Vivo Assessment of Epicardial-derived Cell Migration

Here, we describe methods for isolating primary mouse epicardial cells by an outgrowth culture assay and assessing the functional migration of epicardial-derived cells (EPDC) using an ex vivo heart culture system. These protocols are suitable for identifying genetic and chemical modulators of epicardial epithelial-to-mesenchymal transition (EMT) and motility.
