Anesthetized Animal Injection

Anesthetized animal injection is a neuroscience technique for delivering drugs, tracers, viral vectors, or other substances to an animal while anesthesia limits consciousness and movement, enabling controlled and humane intervention. The procedure typically combines an anesthetic regimen with a sterile syringe or cannula, and may use stereotaxic coordinates to guide a needle to a defined brain region; injection volume, rate, and route determine tissue exposure and distribution. Researchers use this approach to manipulate or label neural circuits, model disease, deliver therapeutics, and measure effects on physiology or behavior. Careful dosing, monitoring, and recovery procedures improve reproducibility and animal welfare.

Injections of AAV Vectors for Optogenetics in Anesthetized and Awake Behaving Non-Human Primate Brain

As currently implemented, optogenetics in non-human primates requires injection of viral vectors into the brain. An optimal injection method should be reliable and, for many applications, capable of targeting individual sites of arbitrary depth that are readily and unambiguously identified in postmortem histology. An injection method with these properties is presented.

Loss of the righting reflex has long served as a standard behavioral surrogate for unconsciousness, also called hypnosis, in laboratory animals. Alterations in volatile anesthetic sensitivity caused by pharmacological interventions can be detected with a carefully controlled high-throughput assessment system, which may be adapted for delivery of any inhaled therapeutic.

Recording Large-scale Neuronal Ensembles with Silicon Probes in the Anesthetized Rat

Extracellular recordings of neuronal activity using silicon probes in the anesthetized rat will be described. This technique allows information to be obtained across multiple brain areas from more than 100 neurons simultaneously. It provides information with single cell resolution about neuronal ensembles dynamics in multiple local circuits.
