Drosophila Brain Immunostaining

Drosophila brain immunostaining is a fluorescence-based technique that labels specific proteins or cellular structures in the fruit fly brain, enabling researchers to visualize nervous system organization and activity-related components. The method typically involves dissecting and fixing brains, permeabilizing tissue so antibodies can enter, and incubating samples with primary antibodies that bind target antigens; fluorescent secondary antibodies then reveal these sites under a microscope. In neuroscience, immunostaining helps map neurons, synapses, neurotransmitter systems, and gene-expression patterns across defined brain regions. Combined with confocal imaging and genetic tools, it supports studies of neural circuits, development, behavior, and disease mechanisms in a genetically tractable model.

Here we describe two assays that have been established to study age-dependent neurodegeneration of dopaminergic (DA) neurons in Drosophila: the climbing/startle-induced negative geotaxis assay which allows to study the functional effects of DA neurons degeneration and the tyrosine hydroxylase immunostaining which is used to identify and count DA neurons in whole brain mounts.

We describe a protocol for dissection, fixation, and immunostaining of steroidogenic organs in Drosophila larvae and adult females to study steroid hormone biosynthesis and its regulatory mechanism. In addition to steroidogenic organs, we visualize the innervation of steroidogenic organs as well as steroidogenic target cells such as germline stem cells.
