Endothelial Cell Permeability

Endothelial cell permeability is the regulated movement of water, ions, solutes, and cells across the endothelial layer lining blood vessels, a property that controls exchange between blood and tissues. It depends on transport through endothelial cells and passage between them, where intercellular junctions, vesicular transport, membrane transporters, and inflammatory signals determine barrier selectivity. In neuroscience, this behavior is central to understanding the blood-brain barrier, whose specialized endothelium limits entry of many circulating substances while permitting essential nutrients to reach neural tissue. Measuring permeability helps investigate neuroinflammation, cerebrovascular disease, drug delivery, and barrier disruption in neurological disorders.

This video demonstrates a protocol for co-culturing mouse brain endothelial cells and primary cortical stem cells and labeling the cell surface glycoproteins using Ac4ManNAz (azidosugar per-O-acetylated N-azidoacetylmannosamine). This method allows for selective enrichment and identification of cell surface proteins in primary cells.

Protein transduction enables the direct delivery of biologically active proteins into cells. In contrast to conventional methods such as DNA transfection or viral transduction this non-invasive paradigm allows highly efficient cellular manipulation in a titratable manner circumventing cellular toxicity and the risk of oncogenic transformation by permanent genetic modification.

The human blood-brain barrier selectively prevents penetration of hydrophilic molecules and pathogens into the brain. Several pathologies, including meningitis and postoperative delirium, are associated with an increased permeability of the blood-brain barrier. Here, we describe an endothelial cell culture model to test the barrier permeability by microbial traversal.

Isolation of Valvular Endothelial Cells

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
