Subretinal Bleb

A subretinal bleb is a localized, fluid-filled separation between the neurosensory retina and retinal pigment epithelium, created to provide access to the outer retina. In a controlled procedure, a fine cannula delivers fluid beneath the retina, producing a temporary detachment that spreads the treatment across a defined area while limiting exposure to surrounding tissue. Subretinal blebs are important in neuroscience and ophthalmic research because they enable targeted delivery of gene therapy vectors, pharmacological agents, or therapeutic cells. Their size, pressure, and duration must be carefully controlled to balance treatment coverage with risks such as retinal injury or impaired visual function.

Subretinal injections are the most common technique for delivering large therapeutic agents such as proteins and viral vectors to photoreceptors and the retinal pigment epithelium. An alternative method in mice that successfully targets the subretinal space with minimal collateral damage and fast recovery times is described here.

Retinal pigment epithelium (RPE) replacement strategies and gene-based therapy are considered for several retinal degenerative conditions. For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.
