Cell Cluster Shape

Cell cluster shape describes the three-dimensional geometry and organization of cells assembled into groups, providing important clues about tissue structure, function, and mechanical behavior. It emerges from coordinated cell-cell adhesion, cytoskeletal tension, polarity, proliferation, and interactions with the surrounding extracellular matrix, which regulate compaction, spreading, and boundary formation. In bioengineering, analyzing cluster shape helps characterize spheroids, organoids, and engineered tissues, while changes in roundness, elongation, or compactness can indicate altered development, disease states, or responses to drugs and biomaterials. Quantifying these features supports the design of more physiologically relevant models for regenerative medicine and therapeutic testing.

A protocol to isolate, culture, and image islet cell clusters (ICCs) derived from human fetal pancreatic cells is described. The method details the steps necessary to generate ICCs from tissue, culture as monolayers or in suspension as aggregates, and image for markers of proliferation and pancreatic cell fate decisions.

The differentiation of stem cells into islet cells provides an alternative solution to conventional diabetes treatment and disease modeling. We describe a detailed stem cell culture protocol that combines a commercial differentiation kit with a previously validated method to aid in producing insulin-secreting, stem cell-derived islets in a dish.

Early development of the fruit fly, Drosophila melanogaster, is characterized by a number of cell shape changes that are well suited for imaging approaches. This article will describe basic tools and methods required for live confocal imaging of Drosophila embryos, and will focus on a cell shape change called cellularization.
