Endothelial Cell Monolayer

An endothelial cell monolayer is a single, continuous layer of endothelial cells that lines blood and lymphatic vessels, forming a selective interface between circulating fluids and surrounding tissues. Its barrier function arises from cell-cell junctions, apical-basal polarity, and regulated transport across the cell layer, while physical cues such as fluid shear stress influence alignment and vascular behavior. In bioengineering, endothelial cell monolayers help model vessel permeability, inflammation, thrombosis, and drug transport in vitro. They also support tissue-engineered vascular grafts, organ-on-a-chip systems, and studies of how engineered materials or biochemical signals affect vascular integrity and function.

Studying Lymphocyte Transmigration Across a Microvascular Endothelial Cell Monolayer

This video demonstrates an experimental investigation of lymphocyte transmigration across a blood-brain barrier (BBB) endothelium model. A monolayer of human brain microvascular endothelial cells (HBMECs) is generated in vitro, and peripheral blood mononuclear cells (PBMCs) containing lymphocytes are added to the monolayer. The cells are then incubated to allow lymphocyte transmigration, and the migrated cells are quantified using flow cytometry.

A Real-time Electrical Impedance Based Technique to Measure Invasion of Endothelial Cell Monolayer by Cancer Cells

This article describes an in vitro technique for monitoring cancer cells invading through a monolayer of endothelial cells. The data is acquired in real-time as a function of changes in impedance on the surface of electrodes at the well bottom.

Intercellular Ca2+-waves are driven by gap junction channels and hemichannels. Here, we describe a method to measure intercellular Ca2+-waves in cell monolayers in response to a local single-cell mechanical stimulus and its application to investigate the properties and regulation of gap junction channels and hemichannels.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.
