Aluminum Sulfate Staining

Aluminum sulfate staining is a biological staining approach in which aluminum sulfate acts primarily as a mordant, helping dyes bind selectively to cellular structures and produce visible contrast. In hematoxylin-based methods, aluminum ions coordinate with oxidized hematoxylin and tissue components to form a stable dye-metal complex, enabling strong nuclear staining after appropriate preparation and washing. This contrast allows researchers and students to distinguish nuclei from surrounding cytoplasm in tissue sections. The technique supports histology, microscopy, and comparative studies of tissue organization, while illustrating how chemical interactions between mordants, dyes, and biological materials determine staining quality and cellular visibility.
