C-fos Immunohistochemical Detection

C-fos immunohistochemical detection is a tissue-labeling method that identifies the c-Fos protein, an immediate early gene product commonly used as an indicator of recent cellular activation. In response to stimuli such as neuronal activity, intracellular signaling induces c-fos transcription, and the resulting c-Fos accumulates in cell nuclei where antibodies can detect it. The procedure typically uses a primary antibody against c-Fos followed by an enzyme-linked or fluorescent secondary antibody, producing a visible signal in tissue sections. In biology, this approach helps map activated neural circuits, relate cellular responses to behavior or pharmacological treatment, and investigate spatial patterns of gene expression.

The use of frozen tissue allows FACS isolation of many brain areas over multiple sessions to maximize the use of valuable animal subjects.
