Cldu Idu Staining

CldU IdU staining is a cell biology method that tracks DNA synthesis by detecting the sequential incorporation of the thymidine analogs chlorodeoxyuridine (CldU) and iododeoxyuridine (IdU) into newly replicated DNA. Cells receive timed CldU and IdU pulses, after which incorporated analogs are detected with antibodies that distinguish their halogenated bases, often in DNA fiber assays or fixed-cell analyses. The resulting labeling patterns reveal replication fork progression, fork speed, origin firing, stalling, and restart. This approach helps researchers assess genome replication dynamics and identify defects associated with DNA damage, replication stress, genetic disorders, and responses to potential therapeutics.

We have derived a strategy to detect sequential incorporation of thymidine analogues (CldU and IdU) into tissues of adult mice to quantify two successive rounds of cell division. This strategy is useful to detect cell turnover of long-lived tissues, oncogenic transformation, or transit-amplifying cells.

In order to examine the cellular, structural and molecular layout of tissues and organs, researchers use a method known as histological staining. In this technique, a tissue of interest is preserved using chemical fixatives and sectioned, or cut into very thin slices. A variety of staining techniques are then applied to provide contrast to the visually uniform sections.
