Dual-channel Fluorescence

Dual-channel fluorescence is an imaging and detection approach that measures signals from two fluorescent labels in the same biological sample, enabling comparison of distinct molecules, structures, or cellular states. Each fluorophore is selected for a different excitation and emission spectrum, while optical filters or separate detector channels isolate the signals and reduce spectral overlap. In biology, this method supports colocalization analysis, live-cell imaging, protein localization, and measurements that use one channel as a reference for the other. Careful control of labeling, exposure, and spectral compensation improves signal interpretation and helps distinguish true biological relationships from bleed-through or background fluorescence.

This protocol describes a general approach to perform photoconversion of fluorescent proteins on a confocal laser scanning microscope. We describe procedures for the photoconversion of puried protein samples, as well as for dual-probe optical highlighting in live cells with mOrange2 and Dronpa.

Dual camera emission splitting systems for two-color fluorescence microscopy generate real-time image sequences with exceptional optical and temporal resolution, a requirement of certain live cell assays including parallel plate flow chamber adhesion assays. When software is employed to merge images from simultaneously acquired emission channels, pseudocolored image sequences are produced.
