Embryonic Larval Zebrafish

Embryonic and larval zebrafish are the early developmental stages of Danio rerio, a vertebrate model that enables direct study of development, physiology, and disease. Because fertilization and embryogenesis occur externally, researchers can observe transparent embryos and larvae by live imaging while manipulating genes, cells, or environmental conditions. These features support investigations of organ formation, nervous-system development, regeneration, toxicology, infection, and drug responses, often with high-throughput experimental designs. Zebrafish studies connect cellular and molecular mechanisms to whole-organism outcomes, providing experimentally accessible insights that can inform vertebrate biology and human health research.

Zebrafish are an emerging system for modeling human disorders of the skeletal muscle. We describe a fast and efficient method to isolate skeletal muscle myofibers from embryonic and larval zebrafish. This method yields a high-density myofiber preparation suitable for study of single skeletal muscle fiber morphology, protein subcellular localization, and muscle physiology.

Forebrain Electrophysiological Recording in Larval Zebrafish

A simple method to record extracellular field potentials in the larval zebrafish forebrain is described. The method provides a robust in vivo read-out of seizure-like activity. This technique can be used with genetically modified zebrafish larvae carrying epilepsy-related genes or seizures evoked by administration of convulsant drugs.

Here we present a protocol to transplant cells with high spatial and temporal resolution in zebrafish embryos and larvae at any stage between at least 1 and 7 days post fertilization.

Development of a Zebrafish Larval Infection Model Using Microgavage

Source: Li, J., et al. Development of a Larval Zebrafish Infection Model for Clostridioides difficile. J. Vis. Exp. (2020).This video demonstrates the microgavage procedure, where an anesthetized zebrafish larva is immobilized in agarose and orally injected with a pathogenic bacterium using a tracer-guided needle. The non-invasive delivery mimics natural infection, enabling analysis of bacterial replication, toxin release, and intestinal epithelial damage.
