Lipid Analysis

Lipid analysis is the identification and measurement of lipids, a diverse group of hydrophobic or amphipathic molecules that support membrane structure, energy storage, and cell signaling. In biological research, samples are typically extracted with organic solvents, separated by properties such as polarity or molecular mass, and characterized using chromatography, mass spectrometry, or targeted biochemical assays. These methods can reveal lipid classes, fatty acid composition, molecular species, and changes in abundance across cells or tissues. Lipid analysis therefore helps investigators study membrane organization, metabolic pathways, signaling responses, and lipid-associated disease, while supporting biomarker discovery and systems-level analysis of cellular function.

Lipid Droplet Isolation for Quantitative Mass Spectrometry Analysis

Lipid droplets are important organelles for the replication of several pathogens, including the Hepatitis C Virus (HCV). We describe a method to isolate lipid droplets for quantitative mass spectrometry of associated proteins; it can be used under a variety of conditions, such as virus infection, environmental stress, or drug treatment.

Lipid polyesters constitute the structural components of two cell wall modifications, the plant cuticle and suberin-containing diffusion barriers. In this video, we describe a method to depolymerize cutin from whole delipidated leaves. The method can be applied to investigating mutants compromised in either cutin or suberin biosynthesis.

This protocol will demonstrate the extraction and analysis of free and esterified bioactive fatty acids from cells. Fatty acids are accurately quantified using stable isotope dilution, chiral liquid chromatography, electron capture atmospheric chemical ionization multiple reaction monitoring mass spectrometry (SID-LC-ECAPCI-MRM/MS).
