Luciferin-luciferase Reagent

Luciferin-luciferase reagent is a chemical system that produces measurable bioluminescence through an enzyme-catalyzed reaction, making it valuable for detecting biological activity. In the presence of appropriate cofactors, luciferase oxidizes luciferin and releases energy as visible light, with the emitted signal reflecting reaction conditions or the amount of reporter enzyme available. In biology, these reagents support reporter gene assays, ATP measurements, enzyme studies, and live-cell or organismal imaging. Because light generation can be highly sensitive and measured without excitation illumination, the system enables quantitative analysis of gene expression, cellular energy status, and biological processes over time.

In this video, we demonstrate a rapid, single-addition assay to assess cell viability in patient-derived organoids upon treatment with a test agent using intracellular ATP levels as a marker for cellular metabolic activity. The intracellular ATP levels are measured using the luciferase/luciferin reaction, where the emitted light intensity indicates the cellular viability and, in turn, the test agent's growth-inhibitory potency.
