Microrna Expression Detection

MicroRNA expression detection measures the abundance of small, noncoding microRNAs in cells, tissues, or biological fluids to reveal post-transcriptional gene regulation. Because mature microRNAs are short and often highly similar in sequence, detection typically involves RNA isolation followed by sequence-specific reverse transcription and quantitative PCR, hybridization-based assays, or small-RNA sequencing. These methods can compare expression across developmental stages, disease states, or experimental treatments while identifying changes in regulatory networks. In biology, microRNA profiling supports studies of cell differentiation, cancer, infection, and biomarker development, linking altered microRNA levels with gene expression patterns and cellular outcomes.

microRNAs are involved in the pathogenesis of IgA nephropathy. We have developed a reliable method for detecting microRNA expression levels in the kidneys of an IgA nephropathy mouse model (HIGA mice). This new method will facilitate to check for miRNAs involvement in IgA nephropathy.

Here we present a protocol for the detection of microRNA expression in rat peritoneal membrane using quantitative real-time reverse-transcription polymerase chain reaction. This method is suitable for studying the microRNA expression profile in rat peritoneal membrane in several pathological conditions.

Quantitative Real Time polymerase chain reaction (qPCR) is a rapid and sensitive method to investigate the expression levels of various microRNA (miRNA) molecules in tumor samples. Using this method expression of hundreds of different miRNA molecules can be amplified, quantified, and analyzed from the same cDNA template.
