Pbs Rinsing Protocol

PBS rinsing protocol is a laboratory procedure that uses phosphate-buffered saline (PBS) to wash cells, tissues, or experimental surfaces while maintaining conditions compatible with biological samples. Because PBS is an aqueous, salt-based buffer designed to remain near physiological pH and osmotic balance, rinsing can remove residual culture medium, loosely associated reagents, or unbound material without exposing samples to abrupt chemical changes. In biology, researchers use this step during cell culture, immunostaining, fixation, and sample preparation to improve cleanliness and reduce background interference, supporting more consistent microscopy, assays, and downstream analyses.

In order to examine gene expression in the pupal wing tissue of Bicyclus anynana, we present an optimized protocol for in situ hybridizations using riboprobes. We also provide guidelines for the further optimization of this protocol for use in pupal wings of other Lepidopteran species.
