Primary Hepatocyte Culture

Primary hepatocyte culture is the ex vivo maintenance of hepatocytes freshly isolated from liver tissue, preserving key features of differentiated liver cells for biological research. After tissue dissociation and cell isolation, hepatocytes are plated on a suitable substrate and maintained in defined culture medium that supports attachment, viability, and liver-specific functions such as metabolism and protein synthesis. This model helps researchers study hepatic physiology, drug metabolism, xenobiotic toxicity, and disease mechanisms under controlled laboratory conditions. Because primary cells more closely reflect native liver behavior than many immortalized cell lines, they provide valuable evidence for pharmacology, toxicology, and regenerative biology.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.
