Sciatic Nerve Transection

Sciatic nerve transection is the complete surgical severing of the sciatic nerve, a major peripheral nerve that controls movement and sensation in the hind limb. Cutting the nerve disrupts axonal continuity and communication between the spinal cord and target muscles, triggering Wallerian degeneration in the distal segment and denervation of affected tissues. In biology, this model is used to investigate peripheral nerve injury, axonal regeneration, neuroinflammation, and functional recovery. Researchers also apply it to evaluate nerve grafts, biomaterials, and regenerative therapies by measuring tissue repair, reinnervation, and restoration of motor or sensory function.

We demonstrate a protocol for axoplasm isolation from adult rat sciatic nerve based on dissection of nerve fascicles and incubation in hypotonic medium to release myelin and lyse non-axonal structures, followed by extraction of the remaining axon-enriched material.

The Sciatic Nerve Cuffing Model of Neuropathic Pain in Mice

Neuropathic pain is a consequence of a lesion or disease affecting the somatosensory system. The “cuff model” of neuropathic pain in mice consists of the implantation of a polyethylene cuff around the main branch of the sciatic nerve. Mechanical allodynia is tested using von Frey filaments.
