Tissue Slicing

Tissue slicing is a biological and histological technique that produces thin sections of organs or other specimens for microscopic examination, preserving cellular and structural relationships. To create a slice, researchers typically fix and embed tissue in paraffin or freeze it, then use a microtome or cryostat to cut sections that can be mounted and stained; precise thickness and orientation influence the quality of the analysis. In biology, tissue slicing supports histology, anatomy, pathology, immunohistochemistry, and studies of spatial organization, enabling researchers to identify cell types, assess tissue changes, and relate microscopic structure to health, disease, and experimental treatment.

Imaging Pheromone Sensing in a Mouse Vomeronasal Acute Tissue Slice Preparation

In mice, the ability to detect pheromones is principally mediated by the vomeronasal organ (VNO). Here, an acute tissue slice preparation of VNO for performing calcium imaging is described. This physiological approach allows observations of subpopulations and/or individual neurons in a living tissue and is convenient for receptor-ligand identification.
