Transgenic Embryo Production

Transgenic embryo production is a biotechnology method for creating embryos that carry an introduced genetic sequence, enabling researchers to study gene function and generate animals with selected traits. The process typically involves inserting DNA into a fertilized egg or early embryo through methods such as pronuclear microinjection, electroporation, or viral delivery; the modified cells then divide as development proceeds. Resulting embryos may be transferred to a surrogate for gestation and screened to identify offspring with stable transgene integration and expression. In biology, this approach supports investigations of development, inheritance, disease mechanisms, and potential agricultural or biomedical applications.

This video demonstrates the ex vivo culture and imaging of oculomotor slices from transgenic mouse embryos. The embryos are isolated and embedded in agarose. Using a vibratome, slices containing the oculomotor nuclei and eyes are obtained. The slices are maintained in culture and imaged using a fluorescence microscope.

The ability to produce transgenes for Caenorhabditis elegans using genomic DNA carried by fosmids is particularly attractive as all of the native regulatory elements are retained. Described is a simple and robust procedure for the production of transgenes via recombineering with the galK selectable marker.

Stably transgenic Hydra are made by microinjection of plasmid DNA into embryos followed by random genomic integration and asexual propagation to establish a uniform line. Transgenic Hydra are used to track cell movements, overexpress genes, study promoter function, or knock down gene expression using RNAi.
