Toxic Compound Exposure

Toxic compound exposure is contact with chemical substances that can disrupt normal biological function, with effects determined by dose, duration, route, and developmental timing. In developmental biology, these compounds may cross protective barriers, alter cellular signaling, damage DNA, or interfere with cell division, migration, differentiation, and tissue formation during embryonic and postnatal development. Researchers study exposure using model organisms, cultured cells, and dose-response experiments to identify developmental toxicity and clarify underlying mechanisms. This work supports risk assessment, environmental health research, and the evaluation of pharmaceuticals, industrial chemicals, and pollutants that may affect reproductive success or produce lasting effects on organismal growth.

Investigation of early endothelial cell (EEC) migration is important to understand the pathophysiology of certain illnesses and to potentially identify novel strategies for therapeutic intervention. The following protocol describes techniques to assess cell migration that have been adapted for the investigation of EEC.

Evaluating the Neuroprotective Effects of a Test Compound Against Alpha-Synuclein Toxicity in Rat Embryonic Neurons

2025

Start with a primary midbrain culture from rat embryos, containing dopaminergic neurons that overexpress mutated alpha-synuclein.Replace the media in the treatment wells with fresh media containing the test compound, and that in the control wells with plain media. Then incubate. In the control, alpha-synuclein forms toxic aggregates in dopaminergic neurons, causing neurite retraction and leading to neuronal death. In the treatment, the test compound interacts with alpha-synuclein, prevents its...

2018

Exposure to environmental toxicants can acutely impact development with long-term effects. Detailed protocols are provided to illustrate a strategy using an effective lab model to study the effect of early embryonic exposure to bisphenol A. We provide fecundity and behavioral assays to monitor the effectiveness of our toxicant exposure bio-assays.

Rapid Evaluation of Toxicity of Chemical Compounds Using Zebrafish Embryos

Zebrafish embryos are used for evaluating the toxicity of chemical compounds. They develop externally and are sensitive to chemicals, allowing detection of subtle phenotypic changes. The experiment only requires a small amount of compound, which is directly added to the plate containing embryos, making the testing system efficient and cost-effective.
