Oocyte Nucleus Spread

Oocyte nucleus spread is a cytogenetic technique that releases and arranges the chromatin from an oocyte nucleus on a microscope slide, enabling direct analysis of meiotic chromosomes. During the procedure, the nuclear envelope is disrupted, nuclear contents are dispersed across the slide, and the material is fixed and stained or labeled for microscopy, preserving chromosome structure and associated proteins. In genetics research, this preparation helps investigators examine chromosome pairing, synaptonemal complex organization, recombination-associated factors, and meiotic abnormalities. It provides a cellular view of chromosome behavior that can clarify mechanisms contributing to aneuploidy, fertility defects, and reproductive disease.

Oogenesis in mammals is known to be error-prone, particularly due to chromosome missegregation. This manuscript describes chromatin spread preparation methods for mouse prophase, metaphase I and II-staged oocytes. These fundamental techniques allow for the study of chromatin-bound proteins and chromosome morphology throughout mammalian oogenesis.

Isolation of Giant Lampbrush Chromosomes from Living Oocytes of Frogs and Salamanders

We present simple techniques for isolating giant transcriptionally active lampbrush chromosomes from living oocytes of frogs and salamanders. We describe how to observe these chromosomes "alive" by phase contrast or differential interference contrast, and how to fix them for fluorescent in situ hybridization or immunofluorescent staining.
