Human Brain Myeloid

Human brain myeloid refers to the immune-cell populations that reside in or enter the central nervous system, including microglia, perivascular macrophages, and infiltrating monocytes. These cells sense tissue damage and infection through pattern-recognition receptors, then coordinate inflammatory signaling, phagocytosis, antigen presentation, and tissue repair. Their activity is shaped by the blood-brain barrier and by communication with neurons, astrocytes, and other immune cells. Studying human brain myeloid cells helps clarify host defense against neuroinvasive pathogens, mechanisms of neuroinflammation, and immune contributions to neurological disease, while informing therapeutic strategies that control harmful inflammation without compromising protection.

Isolation of Myeloid Dendritic Cells and Epithelial Cells from Human Thymus

This protocol details a method to isolate antigen presenting cells from human thymus via different steps of enzymatic digestion of the tissue followed by density centrifugation of the single cell suspension and finally magnetic and/or FACS sorting of the cell populations of interest.

Given the limited xenograft models available to study interactions between human CART and myeloid cells, we established in vitro and in vivo models to understand the impacts of human macrophages on CART cells. Findings can potentially be generalized to evaluate macrophage roles in the tumor microenvironment and test macrophage-targeted immunotherapies.

A Chromatin Assay for Human Brain Tissue

Until recently, expression studies on human brain were limited to quantification of RNA or protein. With the chromatin immunoprecipitation techniques described in this paper, it will be possible to map histone methylation and other epigenetic regulators of gene expression in postmortem brain.
