Monoclonal Antibody Generation

Monoclonal antibody generation is the process of producing large quantities of identical antibodies that bind a single epitope on a defined antigen, enabling precise study and control of immune targets. In a classic hybridoma approach, antibody-producing B cells are isolated after immunization, fused with immortal myeloma cells, and selected in culture; clones are then screened for specificity and expanded to secrete the desired antibody. Recombinant methods can further engineer antibody genes and expression systems to optimize binding, stability, or biological activity. In immunology and infection research, these antibodies support pathogen detection, antigen localization, neutralization studies, diagnostic development, and targeted therapies, while providing standardized tools for investigating host-pathogen interactions.

Generation of Escape Variants of Neutralizing Influenza Virus Monoclonal Antibodies

We describe a method by which we identify critical residues required for the binding of human or murine monoclonal antibodies that target the viral hemagglutinin of influenza A viruses. The protocol can be adapted to other virus surface glycoproteins and their corresponding neutralizing antibodies.

This article provides a detailed protocol for the preparation and evaluation of monoclonal antibodies against natural products for use in various immunoassays. This procedure includes immunization, cell fusion, indirect competitive ELISA for positive clone screening, and monoclonal hybridoma preparation. The specifications for antibody characterization using MALDI-TOF-MS and ELISA analyses are also provided.

Synthesis of human monoclonal antibodies is the first step in studies aimed at unraveling the pathophysiological mechanisms of auto-antibody-mediated immune responses. We have developed a protocol to generate recombinant human immunoglobulin G (IgG) monoclonal antibodies from blood sorted B cells, including B-cell isolation, antibody cloning and in vitro synthesis.

This video demonstrates a method of antibody generation from mammalian cells by introducing plasmid vectors with heavy and light chain genes. These vectors interact with polyethylenimine and enter the cells by forming polyplexes; later, they are translated into individual proteins and assembled into complete antibodies, which are secreted into the extracellular medium.
