Healthy Slice Visualization

Healthy slice visualization is the imaging and examination of tissue slices from non-diseased nervous systems to characterize normal neural structure and organization. Researchers prepare thin slices and use microscopy or related imaging methods to resolve features such as cell bodies, tissue layers, and neuronal connections under controlled conditions. These visualizations establish baseline anatomy and help distinguish normal patterns from changes associated with injury, disease, or experimental treatment. In neuroscience, healthy slices also provide a reference for interpreting tissue-based experiments, validating preparation quality, and linking cellular architecture with neural function.

Visualization and Mapping of Neuronal Pathways in an Amygdala Brain Slice

2025

This video demonstrates a procedure for analyzing synaptic connectivity between medial prefrontal cortex (mPFC) and amygdala neurons using acute amygdala brain slices from mice. The mPFC neurons in these mice express channelrhodopsin fused to a fluorescent protein. The channelrhodopsins facilitate ion influx and action potential generation in the mPFC neurons upon light activation. Consequently, mPFC neurons release neurotransmitters that bind to postsynaptic amygdala neurons, triggering...

Preparing Wedge Brain Slices to Visualize Auditory Neurons

2025

This video demonstrates the process of preparing wedge-shaped brain slices in a laboratory setting. Using a vibratome, a wedge-shaped slice from a mouse's brain is crafted with varying thickness. This slice contains the auditory nerve root positioned on the thicker end, while the hearing-related neurons are located on the thinner end.

Labeling of Biocytin-Filled Interneurons in Rat Hippocampal Slices for Visualization

2025

This video demonstrates the procedure for staining fixed rat brain slices containing biocytin-filled interneurons. The protocol includes incubation with avidin-biotin complex (ABC), staining with HRP and chromogenic substrate, preserving structures with osmium tetroxide, and embedding the tissue in resin for imaging.
