Inhibitory Currents

Inhibitory currents are electrical currents that reduce a neuron’s likelihood of generating an action potential, helping regulate information flow throughout the nervous system. They arise when inhibitory neurotransmitters activate ligand-gated ion channels, commonly permitting chloride ions to enter or potassium ions to leave the cell, which hyperpolarizes the membrane or produces shunting inhibition. In neuroscience, measuring inhibitory postsynaptic currents reveals how synaptic inputs shape neuronal excitability, timing, and network activity. Their balance with excitatory currents supports coordinated signaling and is central to studying neural circuits, sensory processing, learning, and neurological disorders.

A Technique to Assess the Inhibitory Effect of Toxin Exposure on Miniature Excitatory Postsynaptic Currents (mEPSCs)

2025

The video demonstrates the electrophysiological analysis of embryonic stem cell-derived neurons after exposure to neurotoxins. Treating these neurons with botulinum neurotoxins reduces the frequency of spontaneous miniature excitatory post-synaptic currents, confirming inhibition of synaptic transmission.

This paper introduces an approach of combining laser scanning photostimulation with whole cell recordings in transgenic mice expressing GFP in limited inhibitory neuron populations. The technique allows for extensive mapping and quantitative analysis of local synaptic circuits of specific inhibitory cortical neurons.

Investigating Auditory Neuronal Responses in an Awake Mouse in the Presence of an Inhibitory Neurotransmitter Blocker

This video demonstrates the study of neuronal activity in a specific region of the mouse brain that processes auditory stimuli. The technique involves applying an inhibitory neurotransmission blocker and comparing neuronal responses to auditory stimuli before and after the drug is applied to the localized brain region.

A Microtiter Plate Method to Assess the Minimum Inhibitory Concentration of an Antibiotic

2025

This video demonstrates a microtiter plate-based method for determining the Minimum Inhibitory Concentration (MIC) of test antibiotics against bacteria. Upon adding a suspension of the bacterial pathogen Pseudomonas aeruginosa to a microtiter plate containing serial dilutions of a test antibiotic, and an inhibitor of the production of pyomelanin by the bacteria, the optical density of the wells is measured to compute the minimum inhibitory concentration of the antibiotic, and the impact of the...

MicroRNA-Mediated Inhibition of Excitatory Postsynaptic Currents in Mouse Hippocampal Slices

2025

This video demonstrates microRNA-mediated inhibition of excitatory synaptic currents in mouse hippocampal brain slices. The CA3 neurons were infected with a recombinant viral vector harboring a construct for expressing a fluorescent reporter, a light-activated membrane channel, and a microRNA that downregulates the expression of voltage-gated calcium channels. Upon placing a slice in a recording chamber and patching a CA1 neuron with a recording pipette, light pulses were used to stimulate the...
