Acid Pepsin Barrier

The acid-pepsin barrier is a protective system that shields gastrointestinal mucosa from injury caused by gastric hydrochloric acid and the proteolytic enzyme pepsin. It relies on a surface mucus layer containing bicarbonate, intact epithelial tight junctions, rapid cell renewal, and adequate mucosal blood flow to limit acid back-diffusion, neutralize luminal acid near the epithelium, and restrict enzymatic digestion. In pharmacology, this barrier provides a framework for understanding peptic ulcer disease and the actions of acid-suppressing and mucosal-protective drugs. Barrier disruption can promote inflammation, erosions, and ulcers, while strengthening or restoring its components supports mucosal healing.

Improved Method for the Establishment of an In Vitro Blood-Brain Barrier Model Based on Porcine Brain Endothelial Cells

The aim of the protocol is to present an optimized procedure for the establishment of an in vitro blood-brain barrier (BBB) model based on primary porcine brain endothelial cells (pBECs). The model shows high reproducibility, high tightness, and is suitable for studies of transport and intracellular trafficking in drug discovery.

To investigate the blood-retinal barrier permeability and the inner limiting membrane integrity in animal models of retinal disease, we used several adeno-associated virus (AAV) variants as tools to label retinal neurons and glia. Virus mediated reporter gene expression is then used as an indicator of retinal barrier permeability.
