JoVE Neuroscience is a multidisciplinary section devoted to investigations of the structure, function, physiology, and pathophysiology of the brain and nervous system. Included methodologies range from molecular and cellular level studies to full central and peripheral neural systems. JoVE Medicine serves as a connection between basic research and clinical applications. Articles may present medical procedures, case studies or clinical trial methodologies. Included studies are often modeled towards predictive, preventative, personalized, and practical approaches to treatment of disease. JoVE Immunology and Infection includes content from a variety of fields working toward a shared initiative of improving global health. JoVE Engineering encompasses a broad range of experimental and instrumental techniques utilized in physics research. Investigations in this area strive to address and answer a broad range of scientific questions, such as device mechanisms and efficiencies, using physical tools. This approach often requires a combination of specialties, and research in this area tends to be interdisciplinary with contributions from mechanical, electrical, and chemical engineers.

We present a three-color chromatin single molecule localization microscopy (SMLM) staining and analysis protocol that enables reproducible mapping of euchromatin, heterochromatin, and RNAP II for spatial analysis. This protocol enables efficient multicolor labeling in dense nuclear environments, including chromatin-associated targets, allowing reliable simultaneous detection.

A Multilabel Single Molecule Localization Microscopy Protocol for Investigation of Chromatin in the Dense Nuclear Environment

This protocol details the synthesis, hemoglobin encapsulation, and surface PEGylation of hierarchically porous UiO-66 nanoparticles designed for oxygen delivery. The procedure combines defect engineering with strain-promoted biorthogonal conjugation to generate highly permeable and physiologically stable carriers suitable for biomedical applications.

A novel sealing protocol to produce water-tight silicone-based seals for membranes of Polydimethylsiloxane (PDMS) chip devices. Intended for labs setting-up chip models and small-scale chip platform development. We demonstrate the protocol using plastic and native tissue membranes. This procedure only requires PDMS, toluene, mold, membrane, and a vacuum chamber.

Accessible Silicone Chip-to-Membrane Sealing Procedure for Flexible, Reliable Bonding

This protocol describes single-antibody labeling (SAL) to resolve the nanoscale spatial organization of plasma membrane proteins. By leveraging cumulative antibody-epitope interactions at the single-molecule level, membrane SAL (mSAL) maps local epitope distributions while simultaneously capturing antibody binding behavior in the native cellular environment.
