Die Dünnschichtchromatographie (DC) ist eine Technik, mit der organische Verbindungen auf der Grundlage von Polaritätsunterschieden getrennt werden. Ein DC-System besteht aus drei Komponenten: der stationären Phase, dem gelösten Stoff und dem Entwicklungslösungsmittel.
Die stationäre Phase ist die Oberfläche, auf der die Verbindungen getrennt werden, die in der Regel aus extrem polarem Kieselgel besteht. Bei den zu analysierenden Verbindungen oder Gemischen handelt es sich um gelöste Stoffe. Verdünnte Lösungen der gelösten Stoffe werden mit dünnen Kapillaren auf einer Kante der Platte aufgetümmelt.
Der Boden der Platte wird dann in das Entwicklungslösungsmittel getaucht, bei dem es sich in der Regel um ein organisches Lösungsmittel handelt, das weniger polar ist als die stationäre Phase. Das Lösungsmittel wandert durch Kapillarwirkung die Platte hinauf. Wenn das Lösungsmittel durch die Flecken des gelösten Stoffes strömt, trägt es einen Teil des gelösten Stoffes mit sich. Dieses Stoff-Lösungsmittel-Gemisch ist die mobile Phase.
Die Oberseite der mobilen Phase wird als Lösungsmittelfront bezeichnet. Während sich die mobile Phase auf der Platte nach oben bewegt, haften gelöste und lösungsmittelhaltige Moleküle aufgrund ihrer Polarität vorübergehend an der stationären Phase.
Polarere gelöste Stoffe werden stark von der stationären Phase angezogen, so dass sie häufiger haften bleiben und eine Weile stecken bleiben. Weniger polare gelöste Stoffe haften seltener und bleiben nicht lange haften. Das bedeutet, dass weniger polare gelöste Stoffe mehr Zeit in der mobilen Phase verbringen als mehr polare gelöste Stoffe. Mit anderen Worten, weniger polare gelöste Stoffe bewegen sich schneller als mehr polare gelöste Stoffe.
Um eine gute Trennung zwischen den Verbindungen zu erreichen, sollte das Lösungsmittel gerade polar genug sein, um den polarsten gelösten Stoff zu bewegen. Hier sind einige gängige Lösungsmittel in der Reihenfolge zunehmender Polarität. Manchmal werden zwei Lösungsmittel kombiniert, um die gewünschte Polarität zu erreichen.
Da die meisten organischen Verbindungen weiß oder farblos sind, enthalten DC-Platten normalerweise eine fluoreszierende Verbindung. Unter UV-Licht erscheinen die organischen Verbindungen als dunkle Flecken auf der leuchtenden Platte.
Sobald die Verbindungen visualisiert sind, können Sie messen, wie weit sie zurückgelegt haben, und den Verzögerungsfaktor (Rf) für jede einzelne berechnen. Dieser Wert ist die vom gelösten Stoff zurückgelegte Strecke geteilt durch die Gesamtstrecke, die von der mobilen Phase zurückgelegt wurde.
Wenn das Lösungsmittel richtig gewählt wird, hat jeder gelöste Stoff ein deutlich unterschiedliches Rf. Dies ermöglicht es uns, die Komponentenverbindungen in Gemischen zu identifizieren, indem wir die Rf-Werte mit bekannten Verbindungen abgleichen.
In dieser Übung verwenden Sie TLC, um den Rf mehrerer bekannter Verbindungen zu bestimmen, und verwenden dann diese Rf-Werte, um die Identität einer unbekannten Verbindung zu bestimmen.

Bei der herkömmlichen Chromatographie wird die stationäre Phase in eine vertikale Säule gepackt und das Lösungsgemisch am oberen Ende der Säule eingeführt. Wenn das Gemisch durch die stationäre Phase fließt, teilen sich die Verbindungen aufgrund ihrer Strukturen und Polaritäten zwischen der stationären und der mobilen Phase auf und bilden diskrete Banden. Gelöste Stoffe, die schwach mit der stationären Phase wechselwirken, bewegen sich schneller durch die Säule und treten zuerst aus der Säule aus – oder eluieren sie. Gelöste Stoffe, die stärker mit der stationären Phase wechselwirken, bewegen sich langsam durch die Säule und eluieren später. Die Banden können einzeln gesammelt werden, um Verbindungen in einem Gemisch zu isolieren und zu reinigen.
Es gibt verschiedene Arten der Chromatographie, die jeweils eine andere chemische Eigenschaft nutzen, um eine Trennung zu erreichen. Bei der Ionenaustauschchromatographie können beispielsweise die Kügelchen der stationären Phase positiv oder negativ geladen sein und nur Moleküle mit entgegengesetzter Ladung anziehen. Bei der Größenausschlusschromatographie besteht die poröse stationäre Phase aus Kügelchen wie Agarose- oder Dextranpolymeren. Kleinere Moleküle können leichter in die Poren eindringen, während die größeren Moleküle an den Poren vorbeifließen und schneller eluieren.
<h4>Dünnschicht-Chromatographie</h4>
Die Dünnschichtchromatographie (DC) ist eine Art von Chromatographietechnik, bei der Verbindungen anhand ihrer Polarität getrennt werden. Wie bei der herkömmlichen Chromatographie gibt es bei einem DC-System drei Komponenten: die stationäre Phase, die mobile Phase und den gelösten Stoff. Im Gegensatz zur herkömmlichen Chromatographie ist die stationäre Phase jedoch in einer dünnen Schicht auf einer Platte angeordnet und nicht in einer Säule verpackt. Bei der Gleichstromversorgung wird am häufigsten polares Kieselgel, eine Form von Siliziumdioxid, als stationäre Phase verwendet. Die stationäre Phase bildet aufgrund der OH-Gruppen auf ihrer Oberfläche Wasserstoffbrückenbindungen.
Zuerst wird mit einem Bleistift eine Startlinie auf die Unterseite der TLC-Platte gezeichnet. Die zu analysierenden Verbindungen oder Mischungen werden auf der Startlinie mit einer dünnen Kapillare aufgespürt. Dann wird der Boden der Platte in die mobile Phase getaucht, bei der es sich in der Regel um ein organisches Lösungsmittel handelt, das weniger polar ist als die stationäre Phase. Das Lösungsmittel wandert durch Kapillarwirkung die Platte hinauf, passiert die gelösten Flecken und reißt einen Teil jeder Komponente mit sich.
Wenn das Lösungsmittel die Platte hinaufwandert, teilen sich die Komponenten entweder in die mobile Phase oder in die stationäre Phase. Ist das Bauteil polar, interagiert es stärker mit der polaren stationären Phase. Er bewegt sich langsam und bewegt sich nur eine kurze Strecke auf der TLC-Platte. Wenn die Komponente der Probe weniger polar ist – und in der mobilen Phase löslicher ist als in der stationären Phase –, interagiert sie stärker mit der mobilen Phase und bewegt sich weiter auf der DC-Platte. Das Ausmaß der Polarität des Bauteils und die mobile Phase sind essentiell, um die Trennung zu verstehen und vorherzusagen.
DC-Platten enthalten in der Regel einen UV-reaktiven Fluoreszenzfarbstoff, der unter einer UV-Quelle von 254 Nanometern leuchtet. Daher können DC-Platten analysiert werden, indem sie unter UV-Licht betrachtet werden. Verbindungen innerhalb der DC-Platte, wie z. B. die gelösten Stoffe, werden im Vergleich zu einem grünen Hintergrund als dunkle Flecken angezeigt. Indem man die Flecken mit einem Graphitstift umkreist, kann die Strecke gemessen werden, die die Verbindungen relativ zur Lösungsmittelfront zurückgelegt haben. Der Fleck der organischen Verbindung, wenn er nicht selbst fluoreszierend ist, maskiert die Fluoreszenz der Platte und zeigt sich als dunkler Fleck. Einige organische Verbindungen sind UV-aktiv und geben Licht ab, wenn sie UV-Licht ausgesetzt werden. Dabei handelt es sich in der Regel um konjugierte Verbindungen, d. h. solche mit abwechselnden Doppel- und Einfachbindungen, die an der emittierten Wellenlänge zu erkennen sind.
<h4>Verzögerungsfaktor</h4>
Durch die Analyse des Verzögerungsfaktors (Rf) einer Komponente mit einem bestimmten Lösungsmittel kann ein unbekannter gelöster Stoff mit Hilfe von DC bestimmt werden. Der Verzögerungsfaktor ist das Verhältnis der von einer Komponente zurückgelegten Strecke zur von der mobilen Phase zurückgelegten Strecke.

Die von dem gelösten Stoff zurückgelegte Strecke wird von der Startlinie bis zum Mittelpunkt des Spots gemessen, und die von der beweglichen Phase zurückgelegte Strecke wird von derselben Startlinie bis zur Lösungsmittelfront gemessen. Der Verzögerungsfaktor einer Verbindung hängt von der verwendeten mobilen Phase ab. Der Verzögerungsfaktor ist groß für Verbindungen, die hochgradig unpolar sind und eine unpolare bewegliche Phase aufweisen. Niedrige Werte für den Verzögerungsfaktor sind bei polaren Komponenten mit einer unpolaren beweglichen Phase zu beobachten.
Bei einer stark unpolaren mobilen Phase kann es vorkommen, dass sich einige polare Komponenten überhaupt nicht bewegen. Daraus resultiert ein extrem niedriger Verzögerungsfaktor und eine unzureichende Trennung. Eine hochpolare mobile Phase bewirkt, dass sich die Verbindung mit dem Lösungsmittel bewegt und einen extrem hohen Verzögerungsfaktor erzeugt. Dies führt zu einer sehr geringen Trennung zwischen den Komponenten.
Damit die Trennung wirksam ist, sollten die Verzögerungsfaktoren der Komponenten etwa 0,3 - 0,7 auseinander liegen. Um die effiziente mobile Phase zu finden, wird Versuch und Irrtum eingesetzt. Oft erweist sich eine Mischung aus zwei Lösungsmitteln als am effektivsten.

Thin-layer chromatography (TLC) separates compounds in a mixture by polarity. It uses the same basic parts as other chromatography methods: a stationary phase, a mobile phase, and the solute. In TLC, the stationary phase is a thin coating on a plate instead of material packed into a column.

The stationary phase in most TLC systems is polar silica gel, a form of silicon dioxide. Its surface has OH groups that can form hydrogen bonds. The sample is first spotted on a pencil start line with a thin capillary. Then the bottom of the plate is placed in the mobile phase, which is usually an organic solvent that is less polar than the stationary phase.

As the solvent moves up the plate by capillary action, it carries the sample components with it. Each compound then partitions between the mobile phase and the stationary phase. A polar component interacts more with the polar stationary phase, so it moves more slowly and stays near the start. A less polar component is more soluble in the mobile phase, so it travels farther up the plate.

TLC plates often include a UV-reactive fluorescent dye that glows under 254 nm UV light. The compounds appear as dark spots against the green background, and the spots can be circled with a graphite pencil so their movement can be measured. Some organic compounds are UV-active and emit light when exposed to UV light. These are often conjugated compounds, which have alternating double and single bonds, and they can be identified by the wavelength they emit.

The retardation factor, or Rf, helps identify an unknown solute from TLC results. It is the ratio of the distance traveled by the compound to the distance traveled by the solvent front. The compound distance is measured from the start line to the center of the spot, and the solvent distance is measured from the same start line to the solvent front. Rf depends on the mobile phase used.

Rf values are high for highly non-polar compounds with a non-polar mobile phase, and low for polar compounds with a non-polar mobile phase. If the mobile phase is too non-polar, some polar compounds may not move at all. If it is too polar, the compound may move with the solvent and the separation may be poor. For an effective separation, the Rf values of the components should be about 0.3 to 0.7 apart. Choosing the best mobile phase usually takes trial and error, and a mixture of two solvents often works well.
