Rhodamine B Isothiocyanate

Rhodamine B isothiocyanate (RITC) is a red-orange fluorescent labeling reagent used to visualize and track biomolecules, cells, and engineered materials. Its reactive isothiocyanate group forms a covalent thiourea bond with primary amines, allowing attachment to proteins, antibodies, peptides, or other amine-containing substrates while retaining rhodamine fluorescence. In bioengineering, RITC labeling supports fluorescence microscopy, molecular tracking, particle characterization, and evaluation of biomaterial interactions with cells. Because labeled structures can be detected without changing their physical location, the reagent helps researchers analyze transport, distribution, binding, and cellular uptake in engineered biological systems.

Labeling F-actin Barbed Ends with Rhodamine-actin in Permeabilized Neuronal Growth Cones

A method to visualize and quantify F-actin barbed ends in neuronal growth cones is described. After culturing neurons on glass coverslips, cells are permeabilized with a saponin-containing solution. Then, a short incubation with the saponin buffer containing rhodamine-actin incorporates fluorescent actin onto free actin barbed ends.

In the present study, fluorescein-isothiocyanate-labeled (FITC) dextran is administered to mice via oral gavage to evaluate intestinal permeability both in vivo and in plasma and fecal samples. As gut barrier function is affected in many disease processes, this direct and quantitative assay can be used in diverse research areas.

Here, we present a protocol to test glomerular permeability in mice using a highly sensitive, nonradioactive tracer. This method allows repetitive urine analyses with small urine volumes.

Here, we present a protocol for studying mating competitiveness of male Aedes aegypti using fluorescent dye as a marker. Female mosquitoes are exposed to both marked and unmarked males for copulation. Post mating, their spermathecae are examined under a fluorescence microscope to determine their mating partner.
