Beta-catenin Gain-of-function

Beta-catenin gain-of-function refers to increased or constitutive activity of beta-catenin, a protein that regulates cell adhesion and Wnt-dependent gene expression. Normally, the destruction complex phosphorylates cytoplasmic beta-catenin and targets it for degradation; activating mutations or altered signaling can prevent this process, allowing beta-catenin to accumulate, enter the nucleus, and activate TCF/LEF target genes. This mechanism influences cell fate, proliferation, and tissue patterning during development. Studying beta-catenin gain-of-function helps researchers model developmental abnormalities, investigate tumor formation, and evaluate how dysregulated Wnt signaling contributes to disease.
