Oil Shear T-junction

An oil shear T-junction is a microfluidic device that generates uniform droplets by directing an aqueous phase into an immiscible oil stream, enabling precise compartmentalization of biological materials. At the junction, the flowing oil applies shear to the aqueous phase, while interfacial tension and controlled flow rates cause it to pinch off into discrete droplets. These droplets can encapsulate cells, proteins, nucleic acids, or reagents under small-volume, reproducible conditions. In biology, oil shear T-junctions support high-throughput single-cell analysis, digital assays, chemical screening, and studies requiring isolated reaction environments, improving experimental control while reducing sample and reagent consumption.

Different levels and patterns of fluid shear are known to modulate endothelial gene expression, phenotype and susceptibility to disease. We discuss the assembly and use of 'shear rings': a model that produces unidirectional, periodic shear stress patterns. Shear rings are simple to assemble, economical and can produce high cell yields.
