Osteogenic Adipogenic Chondrogenic Differentiation

Osteogenic, adipogenic, and chondrogenic differentiation refers to the directed development of mesenchymal stem or stromal cells into bone-forming, fat-forming, or cartilage-forming lineages, respectively. In vitro, researchers guide these fates by exposing cells to lineage-specific culture conditions containing defined nutrients, signaling molecules, and extracellular matrix cues that activate distinct transcriptional programs. Resulting cells can be evaluated through changes in morphology, matrix production, and lineage-associated markers, such as mineral deposition, lipid accumulation, or cartilage matrix formation. These assays help characterize stem cell potency, compare biomaterials and culture systems, and support research in tissue engineering, regenerative medicine, development, and disease modeling.

Mechanical stress can induce the chondrogenic differentiation of stem cells, providing a potential therapeutic approach for the repair of impaired cartilage. We present a protocol to induce the chondrogenic differentiation of adipose-derived stem cells (ASCs) using centrifugal gravity (CG). CG-induced upregulation of SOX9 results in the development of chondrogenic phenotypes.

Quantitative measurement of bone progenitor function in fracture healing requires high resolution serial imaging technology. Here, protocols are provided for using intravital microscopy and osteo-lineage tracking to sequentially image and quantify the migration, proliferation and differentiation of endogenous osteogenic stem/progenitor cells in the process of repairing bone fracture.

Fibro-Adipogenic Progenitor Isolation, Expansion, and Differentiation from the Spiny Mouse Model

2024

This protocol outlines the isolation of skeletal and cardiac muscle fibro-adipogenic progenitors (FAPs) from spiny mouse (Acomys) via enzymatic dissociation and fluorescence-activated cell sorting. The FAPs obtained from this protocol can be effectively expanded and differentiated to myofibroblasts and adipocytes.

Traditional methods of assessing adipogenic differentiation are cheap and easy to use, but are not specific to changes in gene expression. We have developed an assay to quantify mesenchymal cell differentiation into mature adipocytes using a lineage specific marker. This assay has diverse applications across basic research and clinical medicine.
