Synaptic Transmission

Synaptic transmission is the process by which neurons communicate with other neurons, muscles, or glands, making it essential for nervous system function and behavior. When an action potential reaches a presynaptic terminal, voltage-gated calcium channels open, triggering synaptic vesicles to release neurotransmitters into the synaptic cleft; these molecules bind receptors on the postsynaptic cell and alter ion flow, producing excitatory or inhibitory responses. Synaptic transmission enables sensory processing, movement, learning, and memory, while its disruption contributes to neurological and psychiatric disorders. Studying this process supports research into brain development, neural circuits, and targeted therapeutics.

Paradigms for Pharmacological Characterization of C. elegans Synaptic Transmission Mutants

This video demonstrates how to employ two neural stimulants, aldicarb and pentylenetetrazole (PTZ), in complementary ways to study synaptic function in the nematode, C. elegans. This complementary approach may also be used to shed light on evolutionarily conserved mechanisms for modulating neuronal synchrony and has implications for epilepsy and seizures.

Vibrodissociation of Neurons from Rodent Brain Slices to Study Synaptic Transmission and Image Presynaptic Terminals

This report demonstrates a technique for mechanical isolation of individual viable neurons retaining attached presynaptic boutons. Vibrodissociated neurons have the advantages of rapid production, excellent pharmacological control and improved space-clamp without influence from neighboring cells. This method can be used for imaging of synaptic elements and patch-clamp recording.

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

Non-invasive Assessment of Changes in Corticomotoneuronal Transmission in Humans

The aim of the present study was to assess changes in transmission at the corticomotoneuronal synapses in humans after repetitive transcranial magnetic stimulation. For this purpose, an electrophysiological method is introduced that allows assessment of pathway specific corticospinal transmission, i.e. differentiation of fast, direct corticospinal pathways from polysynaptic connections.
