Vascular Regeneration

Vascular regeneration is the biological restoration or formation of blood vessels after injury, disease, or developmental change, helping reestablish tissue perfusion and oxygen delivery. It depends on coordinated endothelial-cell proliferation, migration, and organization into new vessel structures, often supported by angiogenic signals and interactions with the extracellular matrix and surrounding cells. In biology and regenerative medicine, researchers study these processes to repair ischemic tissues, improve wound healing, and develop engineered grafts or vascularized tissues. Understanding vascular regeneration can also guide therapies for cardiovascular disease and provide models for evaluating how tissues recover their blood supply.

Assessment of Vascular Regeneration in the CNS Using the Mouse Retina

The rodent retina has long been recognized as an accessible window to the brain. In this technical paper we provide a protocol that employs the mouse model of oxygen-induced retinopathy to study the mechanisms that lead to failure of vascular regeneration within the central nervous system after ischemic injury. The described system can also be harnessed to explore strategies to promote regrowth of functional blood vessels within the retina and CNS.

Tools used for visualizing vascular regeneration require methods for contrasting the vascular trees. This film demonstrated a delicate injection technique used to achieve optimal contrasting of the vascular trees and illustrate the potential benefits resulting from a detailed analysis of the resulting specimen using µCT and histological serial sections.

In this work, we present a technique for the rapid fabrication of living vascular tissues by direct culturing of collagen, smooth muscle cells and endothelial cells. In addition, a new protocol for the mechanical characterization of engineered vascular tissues is described.

A System for Culturing Iris Pigment Epithelial Cells to Study Lens Regeneration in Newt

In newt, the lens regenerates always from the dorsal iris by transdifferentiation of the iris pigmented epithelial cells (IPEs). Here we describe a procedure to culture dorsal and ventral newt IPE cells and their implantation to the newt eye. The implanted cells are then studied by tissue sectioning and immunohistochemistry.
