Z-stack Reconstruction

Z-stack reconstruction is an imaging method that combines a series of optical sections collected at different depths into a three-dimensional representation of a specimen. During acquisition, a microscope records sequential focal planes through a defined depth, and reconstruction software aligns these images to visualize cellular structures and spatial relationships across the volume. In cancer research, this approach helps analyze tumor architecture, cell invasion, vascular organization, and interactions within the tumor microenvironment. By preserving depth information that single-plane images cannot capture, Z-stack reconstruction supports more accurate interpretation of fluorescence microscopy data and enables quantitative studies of cancer progression, treatment response, and tissue organization.

Scalable engineered blood vessels would improve clinical applicability. Using easily sizable 3D-printed guides, rings of vascular smooth muscle were created and stacked into a tubular form, forming a vascular graft. Grafts can be sized to meet the range of human coronary artery dimensions by simply changing the 3D-printed guide size.

Coronoid-Temporalis Pedicled Flap for Orbital Floor Defect Reconstruction

2025

This protocol presents a novel coronoid-temporalis pedicled flap (CTPF) technique for reconstructing small-to-medium orbital floor defects. CTPF avoids microvascular risks while maintaining biomechanical stability. Clinical cases demonstrate effective structural restoration, reduced complications, and preserved facial symmetry, offering a viable alternative to traditional grafts or alloplastic materials.

This article describes a method to obtain a three-dimensional (3D) structure of helically assembled molecules using cryo-electron microscopy. In this protocol, we use HIV-1 capsid assemblies to illustrate the detailed 3D reconstruction procedure for achieving a density map by the iterative helical real-space reconstruction method.
