Single-probe Scms

Single-probe single-cell mass spectrometry (SCMS) is an analytical technique that measures chemical constituents in individual cells, revealing cellular heterogeneity that bulk measurements can obscure. A fine capillary probe samples a small volume from a selected cell, while solvent extraction and electrospray ionization transfer metabolites, lipids, drugs, or other analytes into a mass spectrometer for mass-to-charge analysis. This minimally invasive approach can be applied to living cells and supports studies of metabolism, pharmacological response, cell-to-cell variation, and disease-related chemistry. By linking molecular profiles to individual cells, single-probe SCMS strengthens chemical investigations in biology, medicine, and translational research.

Scanning-probe Single-electron Capacitance Spectroscopy

Scanning-probe single-electron capacitance spectroscopy facilitates the study of single-electron motion in localized subsurface regions. A sensitive charge-detection circuit is incorporated into a cryogenic scanning probe microscope to investigate small systems of dopant atoms beneath the surface of semiconductor samples.

PI(4,5)P2 regulates various cellular functions, but its nanoscale organization in the cell plasma membrane is poorly understood. By labeling PI(4,5)P2 with a dual-color fluorescent probe fused to the Pleckstrin Homology domain, we describe a novel approach to study the PI(4,5)P2 spatial distribution in the plasma membrane at the nanometer scale.

This protocol aims to preserve cell metabolites for single-cell mass spectrometry (SCMS) by combining volatile salt solution washing, rapid liquid nitrogen quenching, freeze-drying, and −8 °C storage. It demonstrates that liquid nitrogen quenching is essential for maintaining metabolite profiles, while prolonged cold storage should be minimized to prevent metabolic alterations.

Hand Controlled Manipulation of Single Molecules via a Scanning Probe Microscope with a 3D Virtual Reality Interface

We demonstrate the precise manipulation of individual organic molecules on a metal surface with the tip of a scanning probe microscope driven in 3D by the experimenter's hand using a motion capture system and fully immersive virtual reality goggles.
