Functional Rpe Maturation

Functional RPE maturation is the developmental process by which retinal pigment epithelial cells acquire the specialized structure and activities needed to support photoreceptors and maintain retinal homeostasis. During maturation, RPE cells become polarized, establish tight junctions and epithelial barrier properties, develop apical and basal transport functions, and gain the ability to phagocytose photoreceptor outer segments and participate in the visual cycle. In developmental biology, these functional endpoints help distinguish cell identity from true tissue maturation in stem-cell-derived RPE models. This process supports research on retinal development, disease mechanisms, drug testing, and cell-based therapies for degenerative eye disorders.

Dissection of Human Retina and RPE-Choroid for Proteomic Analysis

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
