Plasma Proteomics

Plasma proteomics is the large-scale study of proteins circulating in blood plasma, providing a molecular view of physiology, disease, and genetic variation. In typical workflows, plasma proteins are separated or enriched, digested into peptides, and identified and quantified by liquid chromatography coupled with mass spectrometry; affinity-based assays can measure selected proteins with high sensitivity. In genetics, these protein profiles help connect genetic variants to molecular phenotypes, reveal protein quantitative trait loci, and clarify pathways linking genotype to disease. Plasma proteomics also supports biomarker discovery, patient stratification, and evaluation of how inherited variation influences treatment response.

Skeletal Muscle Gender Dimorphism from Proteomics

A straight-forward set of methods to isolate and determine the identity of the most abundant proteins expressed in skeletal muscle. About 800 spots are discerned on a two-dimensional gel from 10 mg muscle; this allows for the determination of gender-specific protein expression. These methods will give equivalent results in most tissues.

Bottom-up and Shotgun Proteomics to Identify a Comprehensive Cochlear Proteome

Proteome analysis of the cochlear sensory epithelium can be challenging due to its small size and because membrane proteins are difficult to isolate and identify. Both membrane and soluble proteins can be identified by combining multiple preparative methods and separation techniques along with high-resolution mass spectrometry.

A Quantitative Glycomics and Proteomics Combined Purification Strategy

A high-throughput protocol was developed for combined proteomics and glycomics purification and LC-MS/MS quantification in plasma. Deamidation analysis of N-linked glycosylation motifs was specific to deglycosylated sites. Accurate quantitation of N-glycans was achieved by coupling filter aided N-glycan separation to the individuality normalization when labeling with glycan hydrazide tags strategy.
