Endothelial Cell Activation

Endothelial cell activation is the process by which vascular endothelial cells shift from a quiescent barrier state to an inflammatory, adhesive, and often more permeable phenotype. In immunology and infection, microbial products and cytokines such as tumor necrosis factor and interleukins trigger signaling pathways that increase adhesion molecules, alter junctional integrity, and promote leukocyte attachment and transmigration. This response helps immune cells reach infected tissue, but excessive or persistent activation can contribute to vascular leakage, thrombosis, and tissue injury. Measuring activation markers and barrier function supports studies of host-pathogen interactions, inflammatory disease, and therapies that modulate vascular inflammation.

The goal of this protocol is to isolate lymphatic endothelial cells lining human lymphatic malformation cyst-like vessels and foreskins using fluorescence-activated cell sorting (FACS). Subsequent cell culturing and expansion of these cells permits a new level of experimental sophistication for genetic, proteomic, functional and cell differentiation studies.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
