Fluorescence Assay

A fluorescence assay is an analytical method that uses light-emitting fluorophores to detect or quantify a target molecule, cellular activity, or biological interaction. When excitation light stimulates the fluorophore, it emits light at a longer wavelength, and the measured intensity reflects the presence or amount of the target under defined conditions. In immunology and infection research, fluorescent labels can identify antibodies, antigens, cytokines, infected cells, or microbial components through binding reactions and fluorescence microscopy or plate-based measurements. These assays support pathogen detection, immune-response profiling, diagnostic development, and evaluation of antimicrobial or immunomodulatory treatments.

(2022)This video demonstrates a high-throughput fluorescent assay to evaluate how gut bacteria affect stress resilience in Caenorhabditis elegans. By tracking blue fluorescence from ruptured gut granules, the assay reveals earlier death signals in worms exposed to test bacteria.

A real-time screening procedure for identifying drugs that interact with G protein-gated inward rectifier K+ (GIRK) channels is described. The assay utilizes membrane potential-sensitive fluorescent dyes to measure GIRK channel activity. This technique is adaptable for use on a number of cell lines.

An efficient method to assess surface-exposure of leptospiral proteins is described. The method is specifically designed to avoid disruption of the fragile outer membrane of leptospiral cells. This technique requires employment of several negative controls to assess the integrity of the outer membrane and specificity of antibody reaction.

The zebrafish is a popular tool to model chronic kidney disease (CKD). However, their small size makes it impossible to evaluate renal function using traditional methods. We describe a fluorescent dye kidney clearance assay1 that allows quantitative analysis of zebrafish kidney function in CKD.
