Native Evs

Native EVs are naturally produced extracellular vesicles, including heterogeneous membrane-bound particles released by healthy, infected, or immune cells, that mediate communication between cells. They form through endosomal pathways or outward budding of the plasma membrane, enclosing selected proteins, lipids, and nucleic acids that can be protected during transport and delivered to recipient cells through binding, fusion, or internalization. In immunology and infection, native EVs help regulate antigen presentation, inflammation, innate and adaptive immune responses, and interactions between pathogens and host cells. Their molecular cargo and distribution also support research into disease mechanisms, minimally invasive biomarkers, and vesicle-based therapeutic strategies.

The therapeutic application of extracellular vesicles (EVs) has the potential to revolutionize cancer treatment and drug delivery. Chimeric antigen receptor (CAR) cell-derived EVs (CAR-EVs) isolated using ion-exchange chromatography exhibit increased cargo capacity, significantly enhancing their functional efficacy. This study further characterizes CAR-EVs to elucidate their biological activity and therapeutic potential.

In this video, we demonstrate the SDS-PAGE in-gel digestion method for protein extraction from extracellular vesicles or EVs. Once isolated, the obtained protein fragments can be used for proteomic analysis.

Proteins can either adopt a native structure or misfold into insoluble amyloid. Conditions that favor the misfolding pathway lead to the formation of different types of amyloid fibrils. The methods described here allow rapid conversion of native proteins into amyloid in vitro.

In vitro Synthesis of Native, Fibrous Long Spacing and Segmental Long Spacing Collagen

Simple and reproducible procedures are described for making three structurally distinct collagen assemblies from a common commercially available Type I collagen monomer. Native type, fibrous long spacing or segmental long spacing collagen can be constructed by varying the conditions to which the 300 nm long and 1.4 nm diameter monomer building block is exposed.
