Primary Murine Cells

Primary murine cells are cells isolated directly from mouse tissues and maintained for laboratory study, providing models that more closely reflect normal biology than immortalized cell lines. Researchers typically obtain them through tissue dissection followed by mechanical or enzymatic dissociation, then sustain the resulting populations under defined culture conditions that preserve key cellular functions for a limited period. In immunology and infection research, these cells help characterize immune activation, pathogen entry and replication, inflammatory signaling, and host cell responses. Their physiological relevance supports studies of disease mechanisms and therapeutic effects, while donor variability and limited lifespan require careful experimental design.

The goal of this study was to formulate technologies that allow for successful gene transduction in primary natural killer (NK) cells. The dextran-mediated lentiviral transduction of human or mouse primary NK cells results in higher gene expression efficiencies. This method of gene transduction will vastly improve NK cell genetic manipulation.

Millions of people suffer from retinal degenerative diseases that result in irreversible blindness. A common element of many of these diseases is the loss of retinal ganglion cells (RGCs). This detailed protocol describes the isolation of primary murine RGCs by positive and negative selection with flow cytometry.

Enterochromaffin (EC) cells comprise a small subset of gastrointestinal epithelial cells. EC cells are electrically excitable and release serotonin, yet difficulties in culturing and identifying EC cells have limited physiological studies. The method presented here establishes a primary culture model amenable to examination of single EC cells by electrophysiology.

We describe the rapid isolation of primary murine type II alveolar epithelial cells (AECII) by flow cytometric negative selection. These AECII show high viability and purity and are suitable for a wide range of functional and molecular studies regarding their role in respiratory conditions such as autoimmune or infectious diseases.
