Fucci System

The Fucci system (fluorescent ubiquitination-based cell cycle indicator) is a genetically encoded fluorescence tool that marks cell-cycle phases in living cells, making proliferation and cell-cycle dynamics visible over time. It combines fluorescently tagged fragments of the cell-cycle regulators Cdt1 and Geminin, which are selectively degraded by ubiquitin-mediated proteolysis through phase-specific ubiquitin ligases, producing distinct color signals as cells transition between G1, S, G2, and M phases. In medicine, Fucci supports studies of tumor growth, drug-induced cell-cycle arrest, tissue regeneration, and developmental processes by enabling researchers to track individual cells in cultures, organoids, and animal models. These measurements can clarify treatment responses and heterogeneous proliferation patterns.

The Trowell-type organ culture method has been used to unravel complex signaling networks that govern tooth development and, more recently, for studying regulation involved in stem cells of the continuously growing mouse incisor. Fluorescent-reporter animal models and live-imaging methods facilitate in-depth analyses of dental stem cells and their specific niche microenvironment.

Long-term Live-cell Imaging to Assess Cell Fate in Response to Paclitaxel

Live-cell imaging provides a wealth of information on single cells or whole populations that is unattainable by fixed cell imaging alone. Here, live-cell imaging protocols to assess cell fate decisions following treatment with the anti-mitotic drug paclitaxel are described.

An Electroporation Cytometry Protocol for Live-Cell, Fluorescence Microscopy Using U2 OS Cell Culture

2025

Electroporation is the use of pulsed electric fields (PEFs) to create transient pores in cell cultures to introduce molecular cargo. This technique has recently become widely used in research and clinical settings. Here, we describe electroporation techniques using the recently developed "Electroporation Cytometry System" for performing live-cell, fluorescence microscopy.
