Astrocyte Reprogramming

Astrocyte reprogramming is the conversion of astrocytes, glial cells that support and regulate neural tissue, into neurons or other neural cell types, offering a strategy for studying and potentially repairing damaged nervous systems. The process typically uses defined transcription factors, microRNAs, or changes in cellular signaling to alter gene expression, suppress astrocyte identity, and activate neuronal developmental programs. In neuroscience, researchers use this approach to investigate cell fate, model neurological disease, and generate neurons after injury or during degeneration. Understanding how efficiently and accurately reprogrammed cells integrate into neural circuits is essential for evaluating their therapeutic potential.

This video demonstrates a protocol to evaluate the maturation and reprogramming of glial cells into neurons in a transfected mouse striatum slice. The experiment measures resting membrane potential and response to current injections using neuron-specific fluorescent markers and electrophysiological techniques. Voltage spikes during these measurements indicate successful neuronal maturation and reprogramming.
