Extracellular Neural Signals

Extracellular neural signals are electrical changes generated by neuronal activity and detected outside individual cells, providing a noninvasive view of communication within neural circuits. When neurons fire, ionic currents flow across their membranes and through the surrounding tissue, producing voltage fluctuations that electrodes can record as action potentials or local field potentials. In neuroscience, extracellular recordings help characterize firing patterns, network synchronization, and brain responses to sensory or behavioral events. These measurements support studies of neural coding, disease-related circuit dysfunction, and brain-computer interfaces, while enabling researchers to examine neuronal populations without penetrating every cell.

We demonstrate the fabrication, calibration and properties of two types of ion-selective microelectrodes (double-barreled and concentric) for measurement of ion concentrations in brain tissue. These are then used in the mouse hippocampal slice preparation to show that excitatory activity changes both extracellular potassium and sodium concentrations.

Migraine and its transformation to chronic migraine are immense healthcare burdens in need of improved treatment options. We seek to define how neural immune signaling modulates the susceptibility to migraine, modeled in vitro using spreading depression in hippocampal slice cultures, as a means to develop novel therapeutic targets.
